摘要︰ Oxytocin (OT) ,是在一藉由Ca2+濃度調控模式所分泌的腦垂體後葉激素,其生理功能不但包括子宮的收縮和哺乳,還包含社會行為調控。最近,一些研究顯示腦垂體前葉瘤細胞中,CD38可調節Ca2+濃度變化。因此,作者認為影響OT分泌的關鍵或許就是CD38。首先,作者使用CD38 缺乏(CD38-/-)之母鼠做實驗發現他們的育幼行為被削弱;另外,CD38缺乏之公鼠身上也發現社會身分識別障礙。這兩種行為異常都類似OT 受器缺乏或者OT基因剔除老鼠,而且可被外給OT處理治療。他們也在CD38基因剔除老鼠中發現比起正常鼠的血清和腦脊髓液(CSF)裡找到較低的OT,但是相反結果卻在hypothalamus 和neurohypophysis內觀察到。 那些證據反映出CD38基因剔除老鼠有OT分泌異常的問題,且這個異常可利用CD38再表現的方式恢復。眾所周知細胞內的鈣離子濃度( [Ca2+]i)可被CD38調節,並且OT分泌取決於[Ca2+]i的增加。因此,他們發現藉由阻斷 [Ca2+]i的瞬時增加或CD38的信號路徑能有效減少OT 分泌,以及減少hypothalamic 細胞或者神經末梢的OT分泌就如同CD38缺失的細胞上所發現的情況。總之,他們證明藉由CD38調控Ca2 + 濃度CD38的訊號傳遞路徑是OT分泌的關鍵。 |

In comparison with the wild-type mice, the CD38-deficient mice had significant longer retrieval-time and less duration of crouch, and the defect could be reversed to be normal in CD38-deficient mice after OT treatment. |

A significant higher concentration of OT was noted in the hypothalamus and neurohypophysis. and re-expression of CD38 could also decrease the high levels of OT. |
